Gently invert the tubes 9 times to MIX and immediately place in an ice water bath! MAYBE the original was read as AVOID mixing! hence the confusion!
The use of AFFORD is just a fancy way of saying it!
"Add 250 μl Buffer P2 and invert the tube gently 4–6 times to mix".
If you really want use another word AID is perfectly adequate.
