The coat is padded or quilted; "puffy" means that the padding is thick and this is the "puffiest" winter coat the writer has.

"Serious" is being used here in a common colloquial sense of "very considerable": making "serious" money means making a lot of money.

"Fluff" strictly means "down", that is, soft, voluminous feathers, though it might well be a synthetic equivalent.

"Serious fluff" is a very amusing expression because "fluff" also suggests something light and insubstantial, so "serious fluff" seems paradoxical.

The mention of marshmallow also suggests the possibility that the writer is also playing on the idea of "marshmallow fluff". Marshmallow is a very gooey, glutinous confection and "marshmallow fluff" is a spreadable form of it. Apparently it's very similar to a confection called soapwort meringue (natef) in Arabic cuisine. And it's popular in the northeast of the United States where the winters are very cold and people's fluffiest winter coat would be very thick.
