The coat is padded or quilted; "puffy" means that the padding is thick and this is the "puffiest" winter coat the writer has.

Marshmallow is a very gooey, glutinous confection and "marshmallow fluff" is a spreadable form of it. Apparently it's very similar to a confection called soapwort meringue (natef) in Arabic cuisine. And it's popular in the northeast of the United States where the winters are very cold and people's fluffiest winter coat would be very thick.
