For a start, what is your overall document about? And 'calage' sounds like an activity, so is this about the person doing it, the stage in the production process, etc. etc.?
I think it's necessary to keep the 'chromatique 4 couleurs' together.
I suspect it may in fact be '4-colour registration', but only more context will help us to find out.
'calage' > 'plating' comes direct from Grand Dictionnaire, under 'imprimerie' — so I can't vouch for it personally!

Aha! Then in that case, in the light of the new context, what they seem to be talking about is the "4-colour separations" — or perhaps 'separation' for the name of the activity of producing them; though I must admit I'm more used to hearing it expressed in that way.

Les visuels POS sont d’abord envoyés à CompanyA, qui une fois calé en 4couleurs, envoie leurs fichiers à ZX pour le calage 3couleurs. MR X n’intervient pas dans le process pour les marques américaines.
Le calage chromatique peut prendre beaucoup de temps étant donné que les épreuves & parfois des CD sont envoyés par courrier entre X & Y
