1.Mixing the Chemicals
Prepare solutions before use:
For using your ... it is necessary to prepare 2 chemical solutions. In your box, you will find 2 prepared instant mixtures. You just need to dissolve them in distilled water (which you can normally purchase at your local petrol station).
● Run buffer (RB): add 17 ml of distilled water in the 30 ml tube, put it on the shaker and switch it on. At room temperature it will be completely dissolved in less than 30 min.
● Extraction solution (ES): fill the entire content of the bag into the flask. Dissolve it in roughly 500 ml distilled water by shaking it manually. It will dissolve in less than 1 minute.
1.1.Extraction
When the timer turns to green, you can remove the tube from the shaker and place it in the rack.
Use a syringe to take up roughly 2 ml sample. Now place the filter on the filled syringe. Take a 2 ml vial (small sealed tube) and press the sample through the filter into the vial. The sample should be more or less clear now.

Per utilizzare ... è necessario preparare 2 soluzioni chimiche. Nella scatola troverete 2 miscele istantanee già preparate. È sufficiente scioglierle in acqua distillata (che si può normalmente acquistare presso un distributore di benzina locale).
● Run buffer (RB): aggiungere 17 ml di acqua distillata nel tubo da 30 ml, collocarlo sullo shaker e accendere. A temperatura ambiente si dissolve completamente in meno di 30 min.
● Estrazione della soluzione (ES): riempire l'intero contenuto del sacchetto all’interno della fiala. Scioglierlo in circa 500 ml di acqua distillata agitando manualmente. Si dissolverà in meno di 1 minuto.
1.1.Estrazione
Quando il timer diventa verde, è possibile rimuovere il tubo dallo shaker e metterlo sulla griglia.
Usare una siringa per prelevare circa 2 ml di campione. Posizionare il filtro sulla siringa riempita. Prendere un flaconcino da 2 ml (tubo sigillato piccolo) e premere il campione attraverso il filtro nel flaconcino. Il campione dovrebbe essere ora più o meno chiaro.

I.1.4. Preparazione di campioni e standard
Campioni: pesare circa 0,2 g di tannini enologici sulla bilancia analitica e annotare il peso (corrispondente al peso dell’estratto secco). Disciogliere i tannini enologici in 100 mL di soluzione modello di vino in un matraccio in vetro Pyrex da 100 mL e mescolare bene.
Standard: preparare una soluzione di 10 mg di (-)-epicatechina in 100 mL di acqua distillata corrispondente a una concentrazione di 100 mg/L. Successivamente diluire in acqua distillata fino a ottenere concentrazioni di 0, 20, 40, 60, 80 e 100 mg/L per la curva di calibrazione.
Soluzione modello di vino: preparare una soluzione di 12% vol. di etanolo e 4 g/L di acido tartarico, portare a pH 3,5 con idrossido di sodio.
Soluzione di metilcellulosa: introdurre 300 mL di acqua distillata calda (70-80 °C) in un beker da 1 L e aggiungere lentamente 0,4 g di metilcellulosa. Portare a volume di 1 L con acqua distillata fredda (0 °C). Prima dell’uso, lasciare la soluzione a riposo per una notte a 4 °C.
Soluzione di solfato di ammonio: introdurre 100 mL di acqua distillata calda (30-40 °C) in un beker da 500 mL e aggiungere lentamente 80 g di solfato di ammonio. Portare a volume di 500 mL con acqua distillata. Prima dell’uso, lasciare la soluzione a riposo a temperatura ambiente.

I.1.4. Preparation of samples and standards
Samples: weigh approximately 0.2 g of oenological tannin on the analytical balance and make a note of the weight (corresponding to the dry weight extract). Dissolve the oenological tannin in 100 mL of model wine solution in a 100-mL Pyrex flask and mix well.
Standards: put in solution 10 mg of (-)-epicatechin into 100 mL of distilled water corresponding to 100 mg/L of concentration. Then make dilution in distilled water to obtain 0, 20, 40, 60, 80, and 100 mg/L of concentrations for the calibration curve.
Model wine solution: 12% vol. of ethanol, 4 g/L of tartaric acid adjusted to pH 3.5 with sodium hydroxide.
Methylcellulose solution: put 300 mL of hot distilled water (70-80 °C) in a 1-L beaker and add slowly 0.4 g of methylcellulose. Then fill the beaker until 1 L with cold distilled water (0 °C). Leave the solution before use one night at 4 °C.
Ammonium sulfate solution: put 100 mL of hot distilled water (30-40 °C) in a 500-mL beaker and add slowly 80 g of ammonium sulfate. Then fill the beaker until 500 mL with distilled water. Store the solution at room temperature before use.
